A polymeric multimode waveguide, characterized by a pioneering spoon-shaped geometry and featuring a built-in measuring cell1, was herein proposed to monitor the estradiol in water sample by means a Surface Plasmon Resonance (SPR) biosensor. The fabrication process consists of two steps: a gold deposition on spoon-shaped waveguide polystyrene surface and the functionalization of the sensitive measuring cell with the estrogen receptor (αER) as recognize element. The biosensor response performances were tested in physiological conditions (PBS, 10 mM, pH 7.4) and in different real water matrices of environmental interest (freshwater and seawater). The experimental outcomes supported the possibility to use the αER-based biosensor for the quali-quantitative determination of estradiol in different real water samples in the range of concentration requested by a real scenario without any kind of sample pre-treatment.
